In a social media post, Mayor Jeff Lehman said anyone wishing to leave a message for the families is welcome to do, and the book will be available during business hours for the next several weeks.
Police have said the investigation into the crash will "take time" to complete.
Investigators are concentrating on a large utility hole at the scene of the crash on McKay Road near Veteran's Drive.
Police have said four men and two women who were reported missing on Saturday were believed to be the six people found dead in the crash early Sunday, which had not been reported when officers came across the scene.
The city has said the stretch of McKay Road where the crash happened has been closed to traffic, except local, since the spring due to a municipal construction project.
A vigil for the crash victims is planned for Saturday at the Spirit Catcher on Barrie's waterfront.
